چکیده

Four-day-old C3Hf mice received one daily i.p. injection of phénobarbital (30 mg/kg body weight) in 0.9% NaCI solution or of 0.9% NaCI solution alone for 3 days. On the fourth day the mice were treated with a single i.p. injection of urethan (1 mg/g body weight) in 0.9% NaCI solution or of 0.9% NaCI solution alone. The mice were killed 10 hr after the last injection, and the liver tissue was studied by electron microscopy. Aggregates of an essentially proteinaceous fibrillar mate rial were observed in the hepatocyte nucleoli of 0.9% NaCI solution-treated control mice. After treatment with phéno barbital alone, which induced the typical proliferation of hepatocyte smooth endoplasmic reticulum, the mean num ber of fibrillar aggregates per nucleolus was 19% lower than that of the controls. After treatment with urethan alone, microspherules with a similar structure, but which were somewhat larger and denser than control fibrillar aggregates, were observed. The mean number of microspherules plus fibrillar aggre gates per nucleolus was 91% higher than the mean number of control fibrillar aggregates. After treatment with phéno barbital followed by treatment with urethan, the mean number of microspherules plus fibrillar aggregates per nucleolus was 36% lower than that after treatment with urethan alone. This inhibition of urethan action is probably related to drug-metabolizing enzyme induction by phéno barbital.

Relationship between nucleolar microspherule formation by urethan and inhibition of RNA and protein synthesis.

Liver tissues of suckling C3Hf mice treated with urethan, a carcinogen and inhibitor of RNA synthesis, and with cycloheximide, an inhibitor of protein synthesis, were studied by electron microscopy and by light microscope radioautography. At 4 and 12 hr after treatment with a single dose of urethan (1 mg/g body weight). [3H]uridine incorporation in hepatocyte nuclei was 38 and 9% lower than in ...

Inhibition of mouse hepatocyte gap junctional intercellular communication by phenobarbital correlates with strain-specific hepatocarcinogenesis.

The inhibition of gap junctional intercellular communication (GJIC) is a common effect of nongenotoxic carcinogens and might be a biomarker for these agents. To further test this relationship, we hypothesized that phenobarbital would inhibit mouse hepatocyte GJIC and this would correlate with strain-specific hepatocarcinogenicity. Phenobarbital is a strong nongenotoxic hepatocarcinogen in B6C3F...

P 29: Effects of Different Doses of Some GABAergic Agents in Mouse Brain Under Hypoxic State: Possible Role in Neuro-Inflammation

Introduction: Pro-inflammatory chemokines and cytokines   such as MCP-1 and IL6 can activate microglial cells that has found in some neuro-inflammatory disorders. Hypoxia activates cerebral endothelial cells to release these pro-inflammatory mediators. We aimed to investigate the anti-hypoxic effects of different doses of some GABAergic agents. Materials and Methods: We randomly divid...
